ESTUDIO DE BACTERIAS HALÓFILAS PARA LA PRODUCCIÓN DE BIOMATERIALES TIPO POLIHIDROXIALCANOATOS

Abstract

El objetivo del presente trabajo fue encontrar una cepa halófila productora de PHA por métodos cualitativos y evaluar su rendimiento con distintos parámetros de fermentación a nivel matraz (pH 8, 9, 10 y a 30, 37 y 42oC). La determinación cualitativa fue llevada acabo mediante tinción Azul de nilo y microscopía de fluorescencia, se evaluó la producción de PHA en un medio de cultivo con exceso de sales y el extracto obtenido fue analizado por espectroscopia de infrarrojo (FT-IR). Después de ser evaluada en medio líquido, la producción de biomasa seca resultó en 4.81g/L a 30oC, pH10 y 150rpm. Así como 0.17g/L de PHA a pH 9, 37oC y 150rpm. El análisis FT-IR mostró un pico característico de los PHA cerca de 1700cm-1. La cepa en estudio aislada en el desierto de Cuatrociénegas, Coahuila fue positiva a la tinción específica para producción de PHA. La temperatura de 42oC, pH10 resultó ser la condición para el rendimiento mayor del biopolímero. Los hábitats desérticos son fuente de microorganismos de importancia industrial.
