Abstract

Schizophyllum commune, ülkemizde sıklıkla rastlanan ve tıbbi önemi olan makrofungi türlerinden biri olup, birçok alanda; biyodegredasyon, ilaç sanayi ve gıda endüstrisinde kullanım alanına sahiptir. Çeşitli coğrafik lokasyonlardan elde edilen bu tür canlıların tanımlanması ve biyolojik etkilerinin araştırılması dünya genelinde devam etmektedir. Bu çalışmada, Muğla bölgesinden toplanan bir Schizophyllum türünün identifikasyonu, misel elde edilmesi ve antitümör aktivitelerinin ex vivo açıdan araştırılması hedeflenmiştir. Çalışmada mantar örneğinden misel üretilerek, DNA izolasyonu yapılmış ve ardından ITS gen bölgesi PCR ile çoğaltılarak saflaştırılmıştır. Mantar örneğinin ITS geninin dizi analizi sonucu, S. commune türüne %100 benzerliğe sahip olduğu gösterilmiştir. Ayrıca, mantarın kuru ve misel formlarından elde edilen etanol ekstraktları kullanılarak, PC-3 ve Hela gibi çok yaygın kanser hatlarına karşı sitotoksisitesi araştırılmıştır. Filogenetik analizi de yapılan makrofungi türünün, Beas-2B’ye oranla PC-3 ve Hela kanser hücre hatlarına karşı özellikle 0,25 mg/ml ve altındaki konsantrasyonlarda seçici öldürücü özellik gösterdiği görülmüştür.
