Perubahan Histopatologi Ginjal Tikus Putih yang Diberikan Ekstrak Etanol Sarang Semut dan Gentamisin Dosis Toksik

Abstract

Penelitian ini bertujuan untuk membuktikan bahwa pemberian gentamisin dosis toksik mempengaruhi struktur histopatologis ginjal dan mengetahui efek sarang semut terhadap ginjal tikus putih. Sampel dalam penelitian sebanyak 24 ekor tikus putih jantan yang terdiri dari empat perlakuan yaitu: kontrol negatif (P0) yang diberikan pakan dan minum. Kontrol positif (P1) yang diberikan gentamisin 100 mg/kgBB secara injeksi. Kelompok (P2) diberikan gentamisin 100 mg/kgBB secara injeksi dan ekstrak sarang semut 250 mg/kgBB secara oral. Kelompok (P3) diberikan tujuh hari ekstrak sarang semut dilanjutkan dengan pemberian gentamisin 100 mg/kgBB secara injeksi, dan sarang semut 250 mg/kgBB. Tikus yang telah diberikan perlakuan, kemudian di ambil organ ginjalnya untuk di buat preparat histopatologi. Kemudian dilanjutkan dengan pewarnaan Hematoxylin-Eosin (HE) dan diperiksa di bawah mikroskop dengan pembesaran 400x. variabel yang diperiksa meliputi degenerasi, kongesti, pendarahan, dan nekrosis. Hasil pemeriksaan menunjukkan hasil kerusakan ginjal berupa degenerasi, kongesti, pendarahan, dan nekrosis. Uji Kruskall-Wallis menunjukan adanya perbedaan yang nyata rerata degenerasi, kongesti, pendarahan, dan nekrosis dari kelompok yang diuji. Kesimpulan penelitian ini adalah gentamisin dosis 100 mg/kgBB dapat menyebabkan kerusakan ginjal. Sarang semut dosis 250 mg/kgBB dapat mengurangi kerusakan jaringan.
