Uji Cemaran Mikroba pada Daun Mimba (Azadiractha Indica A. Juss) Sebagai Standarisasi Bahan Obat Herbal

Abstract

Tanaman mimba (Azadiractha indica A. Juss) merupakan bahan herbal yang telah lama digunakan sebagai obat tradisional. Daun mimba mengandung senyawa aktif seperti flavonoid, saponin, tanin, azadiracthin, salanin, meliantriol, nimbin, dan nimbidin yang bermanfaat dalam kesehatan. Penelitian ini bertujuan untuk mengetahui apakah cemaran mikroba yang terdapat pada simplisia daun mimba yang dikoleksi disekitar Univeristas Udayana, Jimbaran, Bali memenuhi persyaratan Badan Pengawasan Obat dan Makanan (BPOM) No. 12 tahun 2014 serta peningkatan mutu dan keamanan obat herbal terstandar. Penentuan cemaran mikroba simplisia daun mimba dilakukan dengan uji angka lempeng total (ALT) mengunakan media Plate Count Agar (PCA) dan angka kapang khamir (AKK) menggunakan media Potato Dextrose Agar (PDA). Pertumbuhan koloni bakteri diinokulasikan pada lempeng agar dengan metode sebar dan diinokulasi pada suhu 35-370C selama 24 -48 jam dan pada suhu 20-250C untuk pertumbuhan angka kapang khamir selama lima hari. Diperoleh hasil dengan pengukuran kuantitaif angka lempeng total dan angka kapang khamir dari simplisia daun mimba yaitu 3750 cfu/g dan 100 cfu/g. Simpulan daun mimba yang dikoleksi dari Kampus Univeristas Udayana, Jimbaran, Bali telah memenuhi persyaratan sesuai dengan standar cemaran mikroba pada obat tradisional pada BPOM RI No. 12 tahun 2014 tentang Persyaratan Mutu Obat Tradisional yaitu jumlah angka lempeng bakteri maksimal 1.000.000 cfu/g dan angka kapang khamir maksimal 10.000 cfu/g dan layak digunakan sebagai bahan baku obat herbal karena telah memenuhi syarat.
