Abstract

В настоящей работе исследована динамика фототоков каналродопсинов-2, экспрессированных в нейронах первичной культуры гиппокампа, в зависимости от частоты, длительности и интенсивности LED источника. Основная цель работы направлена на поиск параметров светового стимула, при которых фототоки не будет значительно изменяться в динамике. Показано, что зависимость между длительностью светового импульса и амплитудой мембранного тока имеет максимум при длительности светового импульса 10-30мс с последующим спадом. Зависимость амплитуды мембранного тока от интенсивности близка к линейной, а зависимость времени достижения максимума от длительности светового стимула по амплитуде достигает насыщения при средних длительностях. Определен оптимальный режим светового стимула, при котором амплитуда мембранных фототоков практически не изменялась в динамике. Эти сведения будут полезны при исследовании нейронов в норме и при патологии, т.к. позволят определить вклад системы каналов ChR2 и выделить искомые различия в норме и при патологии.

In the present work was investigated the dynamics of membrane currents induced by activation of channelrhodopsin-2 (ChR2) expressed in hippocampal neurons in culture. The main goal of the work was to find parameters for the light stimulus (its intensity and duration). It was shown that the dependence of the current amplitude on the light pulse duration at normal conditions is described by a right-skewed bell-shaped curve, while the dependence on the stimulus intensity is close to linear; the time duration is required to reach. Thus, was identified the optimum values ​​of range for the pulse duration (10-30 ms) and the intensity (maximal values), which keep the above stability. The results of certain caveats for discovering pathologies in the activity of neurons: the case, when such dynamics loses the stability of the revealed optimal regime, it may indicate the presence of anomalous neuronal states and e.g. developing neurodegenerative diseases.
