Abstract

Данная работа посвящена разработке технологических этапов культивирования для производства моноклонального антитела цетуксимаб. В рамках проекта рассмотрены подготовительные стадии, получение посевного материала и процессы продуктового культивирования. Основные задачи исследования: 1. Анализ существующей технологии производства рекомбинантного моноклонального антитела (на примере экулизумаба). 2. Организация трансфера технологии с детальным описанием этапов культивирования, включая: o Приготовление питательной среды, o Подготовку посевного материала, o Культивирование в осевом биореакторе. 3. Оценка безопасности разрабатываемых технологических процессов. Объект исследования — проект планировки производственного участка на базе действующего биотехнологического предприятия. Проектирование выполнено на основе методик отдела главного биотехнолога, специализирующегося на разработке технологий производства активных фармацевтических субстанций. Структура дипломного проекта включает: - Описание готового препарата (характеристики, назначение); - Обоснование выбора штамма-продуцента; - Подбор и анализ технологии производства; -Краткую характеристику производственной площадки; - Требования к сырью и материалам; - Блок-схему производственного процесса; - Технологические схемы и описание ключевых стадий; - Материальные расчеты этапов; - Подбор оборудования с обоснованием решений; - Оценку безопасности производства. Работа объединяет технические, технологические и экономические аспекты, направленные на создание эффективного и безопасного производства моноклональных антител. При написании литературного обзора использовались следующие базы данных: eLIBRARY.RU, SCOPUS, NCBI. Статистическую обработку экспериментальных данных осуществляли с использованием пакета программ: Microsoft Excel.

This work is devoted to the development of technological stages for the cultivation of cetuximab monoclonal antibody production. The project considers the preparatory stages, obtaining seed material, and the processes of food cultivation. Main research objectives: 1. Analysis of the existing technology for the production of recombinant monoclonal antibodies (on the example of eculizumab). 2. Organization of technology transfer with a detailed description of the cultivation stages, including: -Preparation of the nutrient medium, -Preparation of seed material, -Cultivation in an axial bioreactor. 3. Safety assessment of the developed technological processes. The object of research is the project of planning a production site based on an existing biotechnological enterprise. The design is based on the methods of the department of the chief biotechnologist, specializing in the development of technologies for the production of active pharmaceutical substances. The structure of the diploma project includes: - Description of the finished product (characteristics, purpose); -Justification for the selection of the producer strain; - Selection and analysis of production technology; - Brief description of the production site; - Requirements for raw materials and materials; - Flowchart of the production process; - Flow charts and description of key stages; - Material calculations of stages; - Selection of equipment with justification of decisions; - Production safety assessment. The work combines technical, technological, and economic aspects aimed at creating an effective and safe production of monoclonal antibodies. When writing the literature review, the following databases were used: eLibrary.RU, SCOPUS, NCBI. Statistical processing of experimental data was carried out using the software package: Microsoft Excel.
