Abstract

Целью данной работы является оптимизация свойств штамма живой гриппозной вакцины, созданного методом обратной генетики, в соответствии с критериями безопасности и высокой репродуктивности в развивающихся куриных эмбрионах. Реассортантный штамм получен методом обратной генетики на основе эпидемического штамма А/Canberra/1/2022(H3N2), и универсального высокорепродуктивного донора A/Leningrad/134/17/57 (H2N2)C. Репродуктивная активность штамма в системе развивающихся куриных эмбрионов и стабильной формулой генома 6:2 с гемагглютинином от штамма дикого типа была поднята до необходимого уровня для производства вакцин. Фенотипические свойства полученного RG штамма соответствуют необходимым для разработки живой гриппозной вакцины. Штамм обладает аттенуирующими мутациями, соответствующими донору аттенуации А/Leningrad/134/17/57 (H2N2), а так же приобрел мутации характерные для развития в клетках птиц.

The purpose of this work is to optimize the properties of a strain of live influenza vaccine created by the method of reverse genetics, in accordance with the criteria of safety and high reproduction in developing chicken embryos. The reassortant strain was obtained by reverse genetics based on the epidemic strain A/Canberra/1/2022(H3N2), and the universal highly productive donor A/Leningrad/134/17/57 (H2N2)C. The reproductive activity of the strain in the system of developing chicken embryos and the stable 6:2 genome formula with hemagglutinin from the wild type strain was raised to the required level for the production of vaccines. The phenotypic properties of the obtained RG strain correspond to those necessary for the development of a live influenza vaccine. The strain has attenuating mutations corresponding to the attenuation donor A/Leningrad/134/17/57 (H2N2), as well as acquired mutations characteristic of development in bird cells.
