Целью данной работы ÑвлÑетÑÑ Ð¾Ð¿Ñ‚Ð¸Ð¼Ð¸Ð·Ð°Ñ†Ð¸Ñ ÑвойÑтв штамма живой гриппозной вакцины, Ñозданного методом обратной генетики, в ÑоответÑтвии Ñ ÐºÑ€Ð¸Ñ‚ÐµÑ€Ð¸Ñми безопаÑноÑти и выÑокой репродуктивноÑти в развивающихÑÑ ÐºÑƒÑ€Ð¸Ð½Ñ‹Ñ… Ñмбрионах. РеаÑÑортантный штамм получен методом обратной генетики на оÑнове ÑпидемичеÑкого штамма Ð/Canberra/1/2022(H3N2), и универÑального выÑокорепродуктивного донора A/Leningrad/134/17/57 (H2N2)C. Ð ÐµÐ¿Ñ€Ð¾Ð´ÑƒÐºÑ‚Ð¸Ð²Ð½Ð°Ñ Ð°ÐºÑ‚Ð¸Ð²Ð½Ð¾Ñть штамма в ÑиÑтеме развивающихÑÑ ÐºÑƒÑ€Ð¸Ð½Ñ‹Ñ… Ñмбрионов и Ñтабильной формулой генома 6:2 Ñ Ð³ÐµÐ¼Ð°Ð³Ð³Ð»ÑŽÑ‚Ð¸Ð½Ð¸Ð½Ð¾Ð¼ от штамма дикого типа была поднÑта до необходимого ÑƒÑ€Ð¾Ð²Ð½Ñ Ð´Ð»Ñ Ð¿Ñ€Ð¾Ð¸Ð·Ð²Ð¾Ð´Ñтва вакцин. ФенотипичеÑкие ÑвойÑтва полученного RG штамма ÑоответÑтвуют необходимым Ð´Ð»Ñ Ñ€Ð°Ð·Ñ€Ð°Ð±Ð¾Ñ‚ÐºÐ¸ живой гриппозной вакцины. Штамм обладает аттенуирующими мутациÑми, ÑоответÑтвующими донору аттенуации Ð/Leningrad/134/17/57 (H2N2), а так же приобрел мутации характерные Ð´Ð»Ñ Ñ€Ð°Ð·Ð²Ð¸Ñ‚Ð¸Ñ Ð² клетках птиц.
The purpose of this work is to optimize the properties of a strain of live influenza vaccine created by the method of reverse genetics, in accordance with the criteria of safety and high reproduction in developing chicken embryos. The reassortant strain was obtained by reverse genetics based on the epidemic strain A/Canberra/1/2022(H3N2), and the universal highly productive donor A/Leningrad/134/17/57 (H2N2)C. The reproductive activity of the strain in the system of developing chicken embryos and the stable 6:2 genome formula with hemagglutinin from the wild type strain was raised to the required level for the production of vaccines. The phenotypic properties of the obtained RG strain correspond to those necessary for the development of a live influenza vaccine. The strain has attenuating mutations corresponding to the attenuation donor A/Leningrad/134/17/57 (H2N2), as well as acquired mutations characteristic of development in bird cells.
