Abstract

Данная работа посвящена выделению и изучению характеристик антител сыворотки крови человека к гипохлорит-модифицированным липопротеинам низкой плотности. Задачи, решаемые в ходе исследования: 1. Проанализировать литературные источники, в которых рассматривается механизм химической модификации ЛПНП in vivo и роль модифицированных ЛПНП в атерогенезе; 2. Обнаружить антитела к гипохлорит-модифицированным ЛПНП в человеческих сыворотках; 3. Усовершенствовать методику выделения АТ к HOCl-ЛПНП из сыворотки крови человека; 4. Доказать специфичность выделенных АТ к HOCl-ЛПНП; 5. Определить константу диссоциации комплекса антиген-антитело для выделенных АТ к HOCl-ЛПНП. Объекты исследования: сыворотка крови человека, липопротеины низкой плотности, человеческий сывороточный альбумин. В ходе работы в сыворотках крови человека обнаружены антитела к гипохлорит-модифицированным липопротеинам низкой плотности. Антитела к гипохлорит-модифицированным липопротеинам низкой плотности были выделены методом аффинной хроматографии. Методом конкурентного иммуноферментного анализа доказали специфичность связывания выделенных антител с собственным антигеном (гипохлорит-модифицированными липопротеинами низкой плотности). Определена константа диссоциации комплекса антиген-антитело для выделенных антител. Обнаружены две группы антител с различной степенью аффинности. Работа выполнена на базе лаборатории липопротеинов имени академика РАМН А. Н. Климова отдела биохимии ФГБНУ «Институт экспериментальной медицины». При написании литературного обзора были использованы базы данных: NCBI (PubMed), eLIBRARY.RU. Статистическую обработку экспериментальных данных осуществляли с использованием пакета программ Microsoft Office (Microsoft Excel).

This work is devoted to the isolation and study of the characteristics of human serum antibodies to low-density lipoproteins modified with hypochlorite. Tasks that were solved during the study: 1. Analyze literature sources that discuss the mechanism of chemical modification of LDL in vivo and the role of modified LDL in atherogenesis; 2. Detect antibodies to hypochlorite-modified LDL in human sera; 3. Improve the method for isolating antibodies to HOCl-LDL from human serum; 4. Prove the specificity of the isolated antibodies to HOCl-LDL; 5. Determine the dissociation constant of the antigen-antibody complex for the isolated antibodies to HOCl-LDL. Object of study: human blood serum, low-density lipoproteins, human serum albumin. During the work, antibodies to hypochlorite-modified low-density lipoproteins were detected in human blood sera. Antibodies to hypochlorite-modified low-density lipoproteins were isolated by affinity chromatography. Using the competitive enzyme immunoassay method, the specificity of binding of isolated antibodies to their own antigen (hypochlorite-modified low-density lipoproteins) was proven. The dissociation constant of the antigen-antibody complex for the isolated antibodies was determined. Two groups of antibodies with different degrees of affinity were discovered. This work was carried out based on the Laboratory of Lipoproteins named after Academician of the Russian Academy of Medical Sciences A.N. Klimov, Department of Biochemistry, Institute of Experimental Medicine. When writing the literature review, the following databases were used: NCBI (PubMed), eLIBRARY.RU. Statistical processing of experimental data was carried out using the Microsoft Office software package (Microsoft Excel).
