Abstract

При написание литературного обзора использовались базы данных: Кибер-Ленинка, ЭБС Айбукс.ру, elib.spbstu.ru, eLIBRARY.RU, PudMed. Статистическую обработку экспериментальных данных осуществляли с ис-пользованием пакета программ: Microsoft Excel. Тема выпускной квалификационной работы: Культивирование штамма-продуцента E. coli с целью снижения содержания бактериальных эндотоксинов. Цель работы: разработать проблему в работе с E. coli – образование эндо-токсинов и методы уменьшения их концентрации. Задачи исследования: - провести обзор литературы; - подобрать методики для выполнения исследований; - изучить способы определения эндотоксинов; - найти методы уменьшения содержания эндотоксинов на стадии культивирования клеток E. coli. Объектом исследования является штамм подгруппы E. coli BL21 (DE3). Предметом исследования являются бактериальные эндотоксины, образующиеся в ходе жизненного цикла клеток E. coli. Работа была выполнена в Лаборатории рекомбинантных биотехнологий в СПбНИИВС. Выводы исследований: 1) Использование минеральных сред для культивирования клеток E. coli позволяет уменьшить количество образующихся бактериальных эндотоксинов после культивирования. 2) Изменение солевого состава сред не влияет на количество образующихся бактериальных эндотоксинов. 3) Уменьшение температуры привело к снижению концентрации получен-ной биомассы и увеличило содержание бактериальных эндотоксинов в пробах. 4) Оптимального вариант проведения культивирования клеток E. coli рекомендую использование среды М9 при температуре 37 °С.

When writing the literature review, the following databases were used: Cyber-Leninka, EBS Aibuks.ru, elib.spbstu.ru, eLIBRARY.RU, PudMed. Statistical processing of experimental data was carried out using the software package: Microsoft Excel. Topic of the final qualification work: Cultivation of the producer strain E. coli in order to reduce the content of bacterial endotoxins. The purpose of the work: to develop a problem in working with E. coli – the formation of endotoxins and methods for reducing their concentration. Research objectives: - conduct a literature review; - choose methods for performing research; - learn how to determine endotoxins; - to find methods for reducing the content of endotoxins at the stage of cultiva-tion of E. coli cells. The object of the study is a strain of the subgroup E. coli BL21 (DE3). The subject of the study is bacterial endotoxins formed during the life cycle of E. coli cells. The work was carried out at the Laboratory of Recombinant Biotechnologies at the St. Petersburg Research Institute of Vitality. Research findings: 1) The use of mineral media for culturing E. coli cells can reduce the amount of bacterial endotoxins formed after culturing. 2) Changing the salt composition of the media does not affect the amount of bac-terial endotoxins formed. 3) Decrease in temperature led to a decrease in the concentration of the obtained biomass and increased the content of bacterial endotoxins in the samples. 4) The optimal option for culturing E. coli cells, I recommend using the M9 me-dium at a temperature of 37 °C.
