Abstract

Данная работа посвящена первичному скринингу ряда препаратов на противовирусную активность в отношении респираторно-синцитиального вируса. Для измерения ЭД50 использовали метод «сэндвич» ИФА. Так же, посредством МТТ теста измеряли цитотоксичность (величину ЦТД50) препаратов. Ð˜ÑÑ Ð¾Ð´Ñ из Ð¿Ð¾Ð»ÑƒÑ‡ÐµÐ½Ð½Ñ‹Ñ Ð·Ð½Ð°Ñ‡ÐµÐ½Ð¸Ð¹ рассчитывали Ñ Ð¸Ð¼Ð¸Ð¾Ñ‚ÐµÑ€Ð°Ð¿ÐµÐ²Ñ‚Ð¸Ñ‡ÐµÑÐºÐ¸Ð¹ индекс (ХТИ) для каждого вещества, из ÐºÐ¾Ñ‚Ð¾Ñ€Ñ‹Ñ Ð² последствии отбирали наиболее перспективные соединения. Был проведен скрининг 93 веществ, из Ð½Ð¸Ñ 23 показали ХТИ, допустимый для Ð¿Ð¾Ñ‚ÐµÐ½Ñ†Ð¸Ð°Ð»ÑŒÐ½Ñ‹Ñ Ð¿Ñ€ÐµÐ¿Ð°Ñ€Ð°Ñ‚Ð¾Ð². Самые высокие показатели ХТИ оказались у Ñ‡ÐµÑ‚Ñ‹Ñ€ÐµÑ Ð²ÐµÑ‰ÐµÑÑ‚Ð² - Ð¿Ñ€Ð¾Ð¸Ð·Ð²Ð¾Ð´Ð½Ñ‹Ñ Ð±ÐµÑ€Ð±ÐµÑ€Ð¸Ð½Ð° и борнеолов, которые в дальнейшем планируется исследовать в качестве Ð¿Ð¾Ñ‚ÐµÐ½Ñ†Ð¸Ð°Ð»ÑŒÐ½Ñ‹Ñ Ð»ÐµÐºÐ°Ñ€ÑÑ‚Ð²ÐµÐ½Ð½Ñ‹Ñ Ð¿Ñ€ÐµÐ¿Ð°Ñ€Ð°Ñ‚Ð¾Ð² против РС-вирусной инфекции.

This work is devoted to the primary screening of a number of drugs for antiviral activity against respiratory syncytial virus. The ED50 was measured using the sandwich ELISA method. Also, using the MTT test, the cytotoxicity (CTD50 value) of the drugs was measured. Based on the obtained values, the chemotherapeutic index (CTI) was calculated for each substance, from which the most promising compounds were subsequently selected. 93 substances were screened, of which 23 showed CTI acceptable for potential drugs. The highest CTI values were found in four substances - derivatives of berberine and borneols, which are planned to be investigated in the future as potential drugs against MS-viral infection.
