Abstract

Данная работа посвящена испытаниям прибора для ПЦР-РВ, а именно амплификатора нуклеиновых кислот CFX96, в целях создания рабочего эталона для передачи единицы величины - с более высокого иерархического уровня рабочим средствам измерений, используемым в клинических лабораториях. В ходе проведенных исследований решались следующие задачи: 1. Выбор и описание материала стандартного образца (СО), подбор праймеров и зондов для амплифицируемых последовательностей нуклеотидов; 2. Отработка метода выделения и очистки ДНК; 3. Проведение исследований последовательностей нуклеотидов с использованием систем для цПЦР. Определение метрологических характеристик СО. 4. Проведение исследований системы ПЦР-РВ (CFX96) с использованием СО. 5. Обработка и сравнение результатов, полученных методами цПЦРи ПЦР-РВ. Определение метрологических характеристик рабочего эталона.

This work is devoted to testing the CFX96 nucleic acid amplifier to create a working standard to transfer unit of value from a higher hierarchical level to working measuring tools that are used in clinical laboratories. Research objectives: 1. Selection and characterization of the reference material (SRM), selection of primers and probes for specific sequences of nucleotides; 2. Extraction and purification of DNA; 3. Study of nucleotide sequences using systems of dPCR. Determination of metrological characteristics of the reference material; 4. Testing of the RT-PCR system (CFX96) using a reference material; 5. Processing and comparison of research results (dPCR and RT-PCR re-sults). Determination of the metrological characteristics of the working standard.
