Целью работы ÑвлÑлоÑÑŒ изучение ÑворачиваниÑ/Ñ€Ð°Ð·Ð²Ð¾Ñ€Ð°Ñ‡Ð¸Ð²Ð°Ð½Ð¸Ñ D-глюкоза/D-галактоза-ÑвÑзывающего белка (GGBP) в уÑловиÑÑ Ð¼Ð¾Ð»ÐµÐºÑƒÐ»Ñрного краудинга. Ð”Ð»Ñ ÑÐ¾Ð·Ð´Ð°Ð½Ð¸Ñ ÑƒÑловий краудинга в иÑÑÐ»ÐµÐ´ÑƒÐµÐ¼Ñ‹Ñ Ñ€Ð°ÑÑ‚Ð²Ð¾Ñ€Ð°Ñ Ð±Ñ‹Ð»Ð¸ иÑпользованы краудинг-агенты полиÑтиленгликоль (ПÐГ) и декÑтран 70. Показано, что Ñдвиг Ñередины денатурационного Ð¿ÐµÑ€ÐµÑ Ð¾Ð´Ð° GGBP в Ñторону более выÑÐ¾ÐºÐ¸Ñ ÐºÐ¾Ð½Ñ†ÐµÐ½Ñ‚Ñ€Ð°Ñ†Ð¸Ð¹ GdnHCl по мере ÑƒÐ²ÐµÐ»Ð¸Ñ‡ÐµÐ½Ð¸Ñ ÐºÐ¾Ð½Ñ†ÐµÐ½Ñ‚Ñ€Ð°Ñ†Ð¸Ð¸ краудинг-агентов в иÑÑÐ»ÐµÐ´ÑƒÐµÐ¼Ñ‹Ñ Ñ€Ð°ÑÑ‚Ð²Ð¾Ñ€Ð°Ñ ÑвлÑетÑÑ ÑледÑтвием ÑƒÐ¼ÐµÐ½ÑŒÑˆÐµÐ½Ð¸Ñ Ñвободного объема в раÑÑматриваемой ÑиÑтеме. УÑтановлено, что Ñворачивание белка в выÑÐ¾ÐºÐ¾ÐºÐ¾Ð½Ñ†ÐµÐ½Ñ‚Ñ€Ð¸Ñ€Ð¾Ð²Ð°Ð½Ð½Ñ‹Ñ Ñ€Ð°ÑÑ‚Ð²Ð¾Ñ€Ð°Ñ ÐŸÐГ и декÑтрана ÑопровождаетÑÑ Ð°Ð³Ñ€ÐµÐ³Ð°Ñ†Ð¸ÐµÐ¹ GGBP. При Ñтом ПÐГ ÑпоÑобен медленно деÑтабилизировать белок в незавиÑимоÑти от приÑутÑÑ‚Ð²Ð¸Ñ Ð² раÑтворе денатуранта. ÐÐµÐ¾Ð±Ñ Ð¾Ð´Ð¸Ð¼Ð¾ отметить, что при выÑÐ¾ÐºÐ¸Ñ ÐºÐ¾Ð½Ñ†ÐµÐ½Ñ‚Ñ€Ð°Ñ†Ð¸ÑÑ Ð´ÐµÐºÑтрана Ñтот полимер, по-видимому, может ÑпецифичеÑки взаимодейÑтвовать Ñ GGBP, изменÑÑ Ð¿Ñ€Ð¸ Ñтом конформацию белка.
The aim of this work was to investigate the folding of D-glucose/D-galactose-binding protein (GGBP) in the macromolecular crowding. To simulate the crowding conditions were used crowding agents polyethylene-glycol (PEG) and dextran 70. Shown, that the shift of the middle GGBP denaturation transition towards higher GdnHCl concentrations with increasing concentrations of crowding agents in the solutions is the result of reducing the free volume in the system. It was found, that the protein folding in highly concentrated solutions of dextran and PEG followed by aggregation GGBP. In addition PEG is able to destabilize the protein slowly regardless of the presence of the denaturant in the solution. It should be noted that at high concentrations the dextran, polymer apparently can interact specifically with GGBP, altering the conformation of the protein.
