Abstract

Целью работы являлось изучение сворачивания/разворачивания D-глюкоза/D-галактоза-связывающего белка (GGBP) в ÑƒÑÐ»Ð¾Ð²Ð¸ÑÑ Ð¼Ð¾Ð»ÐµÐºÑƒÐ»ÑÑ€Ð½Ð¾Ð³Ð¾ краудинга. Для создания условий краудинга в Ð¸ÑÑÐ»ÐµÐ´ÑƒÐµÐ¼Ñ‹Ñ Ñ€Ð°ÑÑ‚Ð²Ð¾Ñ€Ð°Ñ Ð±Ñ‹Ð»Ð¸ использованы краудинг-агенты полиэтиленгликоль (ПЭГ) и декстран 70. Показано, что сдвиг середины денатурационного Ð¿ÐµÑ€ÐµÑ Ð¾Ð´Ð° GGBP в сторону более Ð²Ñ‹ÑÐ¾ÐºÐ¸Ñ ÐºÐ¾Ð½Ñ†ÐµÐ½Ñ‚Ñ€Ð°Ñ†Ð¸Ð¹ GdnHCl по мере увеличения концентрации краудинг-агентов в Ð¸ÑÑÐ»ÐµÐ´ÑƒÐµÐ¼Ñ‹Ñ Ñ€Ð°ÑÑ‚Ð²Ð¾Ñ€Ð°Ñ ÑÐ²Ð»ÑÐµÑ‚ÑÑ следствием уменьшения свободного объема в рассматриваемой системе. Установлено, что сворачивание белка в Ð²Ñ‹ÑÐ¾ÐºÐ¾ÐºÐ¾Ð½Ñ†ÐµÐ½Ñ‚Ñ€Ð¸Ñ€Ð¾Ð²Ð°Ð½Ð½Ñ‹Ñ Ñ€Ð°ÑÑ‚Ð²Ð¾Ñ€Ð°Ñ ÐŸÐ­Ð“ и декстрана сопровождается агрегацией GGBP. При этом ПЭГ способен медленно дестабилизировать белок в независимости от присутствия в растворе денатуранта. ÐÐµÐ¾Ð±Ñ Ð¾Ð´Ð¸Ð¼Ð¾ отметить, что при Ð²Ñ‹ÑÐ¾ÐºÐ¸Ñ ÐºÐ¾Ð½Ñ†ÐµÐ½Ñ‚Ñ€Ð°Ñ†Ð¸ÑÑ Ð´ÐµÐºÑÑ‚Ñ€Ð°Ð½Ð° этот полимер, по-видимому, может специфически взаимодействовать с GGBP, изменяя при этом конформацию белка.

The aim of this work was to investigate the folding of D-glucose/D-galactose-binding protein (GGBP) in the macromolecular crowding. To simulate the crowding conditions were used crowding agents polyethylene-glycol (PEG) and dextran 70. Shown, that the shift of the middle GGBP denaturation transition towards higher GdnHCl concentrations with increasing concentrations of crowding agents in the solutions is the result of reducing the free volume in the system. It was found, that the protein folding in highly concentrated solutions of dextran and PEG followed by aggregation GGBP. In addition PEG is able to destabilize the protein slowly regardless of the presence of the denaturant in the solution. It should be noted that at high concentrations the dextran, polymer apparently can interact specifically with GGBP, altering the conformation of the protein.
