Abstract

Роль АТФ-Ð·Ð°Ð²Ð¸ÑÐ¸Ð¼Ñ‹Ñ Ñ„Ð°ÐºÑ‚Ð¾Ñ€Ð¾Ð² в сборке Ñ Ñ€Ð¾Ð¼Ð°Ñ‚Ð¸Ð½Ð° на данный момент изучена Ð¿Ð»Ð¾Ñ Ð¾. Одним из факторов, возможно ÑƒÑ‡Ð°ÑÑ‚Ð²ÑƒÑŽÑ‰Ð¸Ñ Ð² сборке Ñ Ñ€Ð¾Ð¼Ð°Ñ‚Ð¸Ð½Ð°, является фактор ACF. Целью работы являлось изучение влияния мутации в гене субъединицы Acf1 АТФ-зависимого фактора сборки Ñ Ñ€Ð¾Ð¼Ð°Ñ‚Ð¸Ð½Ð° ACF на эффекты, вызываемые действием ионизирующей радиации в раннем эмбриогенезе дрозофилы. С помощью метода сканирующей конфокальной микроскопии оценена продолжительность S-фазы клеточного цикла, полная продолжительность клеточного цикла на этапе раннего эмбриогенеза у эмбрионов дрозофилы линий дикого типа и мутантов по гену Acf1. Также получены данные по гибели ÑÐ¾Ð¼Ð°Ñ‚Ð¸Ñ‡ÐµÑÐºÐ¸Ñ ÑÐ´ÐµÑ€ эмбрионов дрозофилы линий дикого типа и мутантов по гену Acf1. Полученные данные свидетельствуют о сокращении продолжительности S-фазы клеточного цикла у мутантов по гену Acf1, по сравнению с линией дикого типа, без воздействия Ð²Ð½ÐµÑˆÐ½Ð¸Ñ Ð”ÐÐš-Ð¿Ð¾Ð²Ñ€ÐµÐ¶Ð´Ð°ÑŽÑ‰Ð¸Ñ Ñ„Ð°ÐºÑ‚Ð¾Ñ€Ð¾Ð² и после воздействия ионизирующего излучения. Гибель большего числа ядер у Acf1(7)-мутантов после воздействия ионизирующего излучения указывает на участие АТФ-зависимого Ñ Ñ€Ð¾Ð¼Ð°Ñ‚Ð¸Ð½-ассемблирующего фактора ACF в репарации повреждений ДНК на стадии делений дробления.

The role of ATP-dependent chromatin assembly factors in the currently poorly understood. One of the factors, which may participate in the assembly of chromatin, is a factor ACF. The aim of the work was to study the effect of mutations in the gene Acf1 subunit of ATP-dependent chromatin factor ACF assembly on the effects caused by the action of ionizing radiation in the early Drosophila embryogenesis. Using the method of scanning confocal microscope estimated duration of S-phase of the cell cycle, the total duration of the cell cycle at the early stage of embryogenesis in Drosophila embryos of wild-type strains and mutants of the gene Acf1. Also obtained data on the death of somatic nuclei of Drosophila embryos of wild-type strains and mutants by gene Acf1. These data suggest shortening S-phase of the cell cycle in the mutant gene Acf1, compared to the wild-type line, without the influence of external factors, DNA damage after exposure to ionizing radiation. The death of a greater number of cores in Acf1 (7) mutants after exposure to ionizing radiation indicates a part of ATP-dependent chromatin-assembled ACF factor in the repair of DNA damage at the stage of cleavage divisions.
