Abstract

Показано, что первичные культуры мультипотентных мезенхимных стромальных клеток костного мозга, синовиальной ткани и суставных хондроцитов отличались по способности к колониеобразованию, формированию сфероидов, наработке белков внеклеточного матрикса. Полилактидные матрицы не оказывали влияния на жизнеспособность хондроцитов кролика. Оптимальным способом нанесения хондроцитов на полилактидный скаффолд является предобработка его ростовой средой и нанесение клеток в составе коллагенового геля. В условиях индукции хондрогенной дифференцировки МСК костного мозга на полилактидном скаффолде, модифицированном хондроитинсульфатом, происходит активное накопление белков внеклеточного матрикса.

It is shown that primary cultures of the multipotent mesenchymal stromal cells of bone marrow, synovial tissue and articulate chondrocytes differed on ability to form of colonies, spheroids, express of extracellular matrix proteins. Polylactide scaffolds didn't influence on viability of rabbit chondrocytes. An optimal way of settling the chondrocytes on polylactide scaffold is culture medium scaffold processing and drawing cells as a part of collagen gel. Active accumulation of proteins of an extracellular matrix occurs during chondrogenic differentiation of mesenchymal stromal cells of bone marrow, attached to a polylactide scaffold, modified by chondroitin sulfate.
