
ChIP of yeast cells. Exponential phase growing cells are crosslinked, washed and stored at -80°C. Lysis is performed, unsoluble chromatin is isolated, with subsequend sonication of the chromatin to reach an average of 500bp fragments. IP is carried out using magnetic beads. Proteins are degraded, crosslinked is reversed and RNA is degraded. qPCR is performed on purified samples.
