Inmunología de la tripanosomiasis africana por Trypanosoma rhodesiense

Abstract

Los ensayos llevados a cabo en medios de cultivo artificiales para cultivar T rhodesiense, cepa 165, han demostrado que esta cepa, adaptada a los roedores, no es capaz de multiplicarse de forma masiva en estos medios. Por otro lado, la cepa de T.cruzi, mantenida en NNN, no se multiplica en el ratón ni en el cobaya. El suero de conejo inmunizado es superior para detectar los componentes antigénicos de T.rhodesiense que el suero de conejo infestado experimentalmente y que el suero de enfermo con tripanosomiasis. El tratamiento con acetona de los preparados antigénicos ocasiona una pérdida parcial de componentes antigénicos de T.rhodesiense. Los componentes antigénicos hallados en el preparado metabólico obtenido de sangre, o del medio de cultivo libre de células, están también presentes en el extracto antigénico total de T.cruzi y T.rhodesiense. La variabilidad antigénica, a lo largo de pases cortos en ratas Wistar, no es completa. Hay comunidad antigénica parcial entre T.cruzi y T.rhodesiense. Se han revelado varios componentes de málico deshidrogenasa y succínico deshidrogenasa en el enzimoelectroferograma sobre gel de agar, en preparados antigénicos de T.rhodesiense y T.cruzi. La actividad málico deshidrogenasa es mucho mayor que la succínico deshidrogenasa. Las técnicas de aglutinación de tripanosomas vivos y la determinación de IgM por inmunodifusión radial simple han dado resultados favorables en la investigación de la enfermedad del sueño por T.rhodesiense. Por el contrario, debido al número significativo de sueros que presentan anticuerpos heterófilos, la técnica de hemaglutinación indirecta no es útil para el serodiagnóstico de esta enfermedad.
