Variabilidad y diferenciación genética en cuatro poblaciones de la planta medicinal Psychotria acuminata en Costa Rica

Abstract

Un total de 51 accesiones de Psychotria acuminata, provenientes de 4 poblaciones en Costa Rica, fueron evaluadas con los marcadores moleculares RAPD (Random Amplified Polymorphic DNA) e ISSR (Inter-Simple Sequence Repeat DNA). Con el uso de 16 decámeros se obtuvo 70 loci polimórficos y con 3 ISSRs 15 loci polimórficos. Ambos tipos de imprimador fueron capaces de determinar la diversidad genética entre las 4 poblaciones. Los dendrogramas basados en la distancia genética entre las poblaciones no mostraron similitud entre los RAPDs y los ISSRs; sin embargo, ambos métodos revelaron una alta variabilidad genética entre las poblaciones (Gst de 0,292 y 0,313 para los RAPDs y los ISSRs, respectivamente). Estos niveles altos de diversidad genética, entre las 4 poblaciones, no pudieron ser explicados por los factores climáticos (r2=-0,302, P=0,560) ni por las distancias geográficas (r2=-0,183, P=0,768). Con los resultados obtenidos se concluye que las poblaciones de P. acuminata evaluadas presentan un importante nivel de diferenciación genotípica. Como consecuencia, sería posible utilizar la diversidad genética de esta especie no solo en programas de conservación de la biodiversidad, sino también para un mejor uso de sus recursos fitoquímicos.
