Abstract

Dans ce projet de recherche nous avons analyse les glycosides de l'epinette noire. Cette analyse comporte deux parties: 1) - l'analyse des glycosides sans transformation chimique. Les glycosides sont extraits du materiel vegetal sec par solvant. Par des techniques de chromatographie liquide (CCM, CLP et CLHP), quatre glycosides phenoliques sont purifies. Des structures basees sur des donnees spectroscopiques (RMN H1 et 13C et SM) sont proposees. Dans les quatre glycosides, le sucre est le (B-D-glucose. 2) - l'analyse des aglycones des glycosides apres hydrolyse. Une fois les substances volatiles eliminees de l'extrait des glycosides, ce dernier subit une hydrolyse enzymatique pour liberer les aglycones. Deux enzymes sont utilisees pour comparer leur efficacite de l'hydrolyse des glycosides. Les aglycones liberees sont extraites par le pentane et le pentane et l'ether (l:l/v:v), et analysees par chromatographie en phase gazeuse sur deux colonnes capillaires (DB-5 et Supelco-10). La comparaison des pourcentages relatifs des aglycones a revele que la (3-glucosidase est plus efficace pour hydrolyser les glucosides phenoliques et la cellulase est plus efficace pour hydrolyser les glucosides monoterpeniques. Etant donne l'importance des glycosides comme source supplementaire de produits volatils de l'epinette noire, l'objectif de ce travail etait d'evaluer la qualite et la quantite des aglycones des glycosides et d'analyser la structure de certains glycosides par des methodes spectroscopiques.
