Abstract

[ES]Estudio epidemiol?gico caso-control para comprobar la hip?tesis de que ciertos polimorfismos gen?ticos relacionados con las citosinas y con el proceso de autofagia pueden modificar el riesgo de presentar tiroiditis autoinmune. Se realiza el estudio sobre 200 pacientes con tiroiditis autoinmune reclutados en el Servicio de Endocrinolog?a y Nutrici?n en el Complejo Asistencial Universitario de Salamanca, en los que se confirma la ausencia de diferencias en cuanto a caracter?sticas epidemiol?gicas y cl?nicas respecto a los datos habituales poblacionales. Se valora la influencia de 8 polimorfismos gen?ticos, 4 relacionados con citosinas y 4 relacionados con prote?nas del proceso de autofagia. En nuestro estudio no hemos encontrado ninguna asociaci?n significativa entre la presencia de tiroiditis autoinmune y las variantes al?licas y genot?picas de los polimorfismos -511 C>G del gen IL1B, -33T>C del gen IL4, -174 G>C del gen IL6 y -1C>T del gen CD40, lo que muestra que las variantes g?nicas de los genes que codifican prote?nas relacionadas con la respuesta inmune incluidas en nuestro estudio no modifican el riesgo de desarrollar tiroiditis autoinmune. Por primera vez mostramos que ser portador del alelo G (genotipos CG y GG) del polimorfismo -4147 C>G del gen ATG2B (rs3759601) supone un aumento del riesgo de presentar tiroiditis autoinmune. Estos datos sugieren que es posible que las variaciones en el genotipo del gen ATG2B puedan modificar el proceso de autofagia al alterar su funcionalidad, e influir en la etiopatog?nesis de la tiroiditis autoinmune. As? mismo, mostramos por primera vez que ser portador del alelo G (genotipos CG y GG) del polimorfismo 106662741 C>G del gen ATG5 (rs2245214) confiere un efecto protector frente al desarrollo de tiroiditis autoinmune, reforzando la idea de que variaciones en el mecanismo de autofagia pueden estar implicadas en el desarrollo de la enfermedad.
