
Επιλέχθηκαν έξι προελεύσεις καστανιάς (Castanea sativa Mill.) από ακραία περιβάλλοντα της Ελλάδας, της Ιταλίας και της Ισπανίας για να διερευνηθεί η αύξηση και η γενετική παραλλακτικότητα. Μελετήθηκαν 143 οικογένειες ελεύθερης επικονίασης σε ένα πείραμα πεδίου συνδυασμένης δόκιμης προελεύσεων - απογόνων. Η μέση επιβίωση ήταν 69.9% στην ηλικία των έξι ετών. Καταγράφηκε αξιοσημείωτη γενετική παραλλακτικότητα μεταξύ και εντός των προελεύσεων ως προς τα γνωρίσματα αύξησης. Οι αποδοτικότερες προελεύσεις ήταν οι EU-11 (A. Coruna, Ισπανία) για τη διάμετρο, EU-56 (Pellice, Ιταλία) για το ύψος και τον όγκο και EU-7 (Malaga, Ισπανία) για τον αριθμό φύλλων. Παρουσιάζονται οι τιμές των συντελεστών κληρονομικής ικανότητας και αθροιστικής παραλλακτικότητας για τα επιμέρους γνωρίσματα από τις οποίες διαφαίνεται γενικά η ύπαρξη σημαντικής παραλλακτικότητας στο υπό μελέτη γενετικό υλικό. Υποδηλώνεται το σημαντικό δυναμικό του εν λόγω υλικού για την περαιτέρω εφαρμογή προγραμμάτων γενετικής βελτίωσης. Ο μικροπολλαπλασιασμός επιλεγμένου υλικού διερευνήθηκε με ιστοκαλλιέργεια. Χρησιμοποιήθηκαν βλαστοί οκτώ άριστων φαινοτύπων, τρεις συγκεντρώσεις θρεπτικού υποστρώματος και τρεις συγκεντρώσεις κυτοκινίνης (BAP). Στατιστικά σημαντική ήταν η επίδραση του γενοτύπου και του υποστρώματος. Ο συνδυασμός υποστρώματος MS και BAP συγκέντρωσης 0.2 mg/l ήταν ο βέλτιστος στα γνωρίσματα πολλαπλασιασμού που μετρήθηκαν. Επίσης το μεγαλύτερο ποσοστό ριζοβολίας (56%) βρέθηκε στη συγκέντρωση 0.5 mg/l NAA. Τα αποτελέσματα είναι ενδεικτικά των δυνατοτήτων που προσφέρει η ιστοκαλλιέργεια στον πολλαπλασιασμό επιλεγμένου γενετικού υλικού καστανιάς και στην ενσωμάτωση της τεχνικής αυτής στα προγράμματα γενετικής βελτίωσης. Γενικά διαπιστώθηκαν οι μεγάλες δυνατότητες κλασσικής γενετικής βελτίωσης του είδους που είναι πολύ υποσχόμενες σε συνδυασμό με την επιτυχία του μικροπολλαπλασιασμού
The performance and variability of 143 open-pollinated families of Castanea sativa (Mill) from six natural contrasting European chestnut populations (Spain, Greece and Italy) were investigated. The average survival was 69.9% at the end of the sixth year. Considerable genetic variation was recorded both among and within provenances and growth traits were under strong genetic control. The best populations were EU-11 (A. Coruna, Spain) for diameter, EU-56 (Pellice, Italy) for height and volume while EU-7 (Malaga, Spain) for the number of leaves. The heritability as well as CVA were also estimated. Although heritability values can be regarded as potentially over estimated, the large variation observed suggest the possibilities of selecting reproductive materials for tree improvement purposes. For in vitro culture, explants from sprouted buds of eight mature plus-trees of Castanea sativa originating from the two Greek natural populations were cultured. Three nutritional solutions and three growth regulator concentrations (BAP) were used. Results showed high genotypic variation for the number of shoots produced per explant. The highest shoot per explant was obtained with the P-2 genotype with MS medium and 0.2 mg/l BAP being the best on the multiplication traits measured. 56% rooting was achieved in 0.50 mg/l NAA in vermiculite medium after eight weeks. The results indicate that the extensive micro-propagation of selected genotypes is possible and this could be a useful tool for mass propagation of selected plus trees or any other project of tree improvement
