Abstract

現在,うつ病の治療薬はセロトニンやノルアドレナリンといったモノアミンを増加させる薬剤が中心として用いられているが,脳内の細胞外モノアミン濃度は抗うつ薬の投与数時間後には増加するのに対し,臨床における治療効果発現までには数週間の慢性投与が必要とされることもあり,抗うつ薬の治療発現メカニズムは未だ明らかとされていない.近年,成熟期の脳においても海馬歯状回といった特定領域において,神経幹・前駆細胞が存在し,神経細胞が新生されることが明らかにされている.この海馬における神経新生は,うつ病発症の危険因子とされるストレスによって抑制され,逆に抗うつ薬を慢性投与することによって促進される.さらに,海馬神経新生を阻害すると,行動薬理評価モデルにおける抗うつ薬の作用が消失するなどの報告から,抗うつ薬による治療メカニズムの新しい仮説として海馬の神経新生促進仮説が提唱され,注目を集めている.本稿では,うつ病と海馬神経新生の関わりについて概説するとともに,我々が確立した成体ラット海馬歯状回由来神経幹・前駆細胞(Adult rat Dentate gyrus derived neural Precursor cell: ADP)の紹介を交えて,海馬神経新生をターゲットとした新規抗うつ薬創製のアプローチの可能性について記載する.
