The chromatographic behavior of calf thymus histone on different grades of Sephadex with varying degrees of cross-linkages has been studied. Fractions of histone obtained by preliminary separation on carboxymethylcellulose were characterized by chromatography on Sephadex and by electrophoresis on gels of starch and polyacrylamide. Analytical separation on Sephadex has confirmed the ease of fractionation of histones by acid extraction.
