The media and conditions for the growth of isolated maple tissues were determined. Tissues isolated from Acer pennsylvanicum and Acer saccharum were grown on a medium containing 25% coconut milk, 2 p.p.m. naphthalene-acetic acid, 2% sucrose, and 2 times the salt concentrations suggested by White and Nitsch. The cultures have maintained an actively growing condition in the dark at 26–27 °C. The utilization of various carbohydrates by A. pennsylvanicum tissue was determined. Sucrose, glucose, and raffinose were utilized most efficiently. Extracellular starch digestion was detected using isolated A. saccharum stem callus.
