A PREREQUISITE for studying the DNA-protein interactions involved in integration and excision of λ is the determination of the DNA sequences of the attachment sites. Useful sequence data can only be obtained if the position of the attachment site is known precisely. To focus on this site we have used genetically- and physically-defined deletions and substitutions ending in att as reference points whose position relative to the sites of action of restriction endonucleases can be determined. In this way we have located the middle of the attachment site to within ± 10 base pairs in a molecule 46,500 base pairs long1.
