Complete conformational switching in a calcium ionophore

Abstract

A new, conformationally mobile, ether-amide, calcium ionophore, 1(S,R) ,2(S,R),4(R,S)-4-methyl-1,2-bis((N,N-dipropylcarbamyl)methoxy)cyclohexane, is described. The changes in the conformation of the ionophore were monitored by 1 H and 13 C NMR spectroscopy at room temperature and at low temperature (-90 o C). Examination of conformational effects on NMR spectra allowed a direct determination of 2:1 ionophore-to-cation stoi- chiometry and facilitated the measurement of the association constant. Conformational monitoring of the difference in complexing ability at room temperature at -90 o C demonstrated that complexation was more pronounced at room temperature
