Implication de la voie des kynurénines dans la polarisation des macrophages pulmonaires humains

Abstract

Organisation Travail realise sous la direction de P. Devillier. Introduction Les kynurenines (Tranilast®, 3OHAA et 3OHKYN), metabolites du tryptophane sont des molecules immunologiquement actives. Elles jouent un role dans la modulation de la reponse inflammatoire notamment au niveau pulmonaire. Nous avons etudie l’expression de l’indoleamine 2,3 dioxygenase (IDO-1 et 2), premiere enzyme de cette voie, par des macrophages pulmonaires humains puis l’effet des kynurenines sur la polarisation M1/M2 de ces macrophages en fonction de leur spectre de production cytokinique. Methodes A partir de tissus pulmonaires provenant de pieces de resection chirurgicale, nous avons realise des cultures de macrophages isoles par adherence au support. Ces prelevements ont ete stimules par des doses croissantes (200–400 μmol/L) de Tranilast®, 3OHAA ou 3OHKYN en presence ou non de LPS ou d’IL-4. Les macrophages ont egalement ete exposes a des inhibiteurs de l’IDO (LMT/DMT). Apres 24 h de traitement, les cytokines ont ete dosees en ELISA dans les surnageants et nous avons etudie l’expression de la voie en RT-qPCR. Resultats L’indoleamine 2,3 dioxygenase est exprimee par les macrophages pulmonaires humains en reponse au LPS mais pas a l’IL-4. Les kynurenines inhibent la production de cytokines M1 pro-inflammatoires LPS induites (CXCL8, CXCL1, CCL2, TNF-α, CXCL10) ainsi que la production de cytokines M2 (CCL13, CCL22, CCL18) regulatrices IL-4 induites. Paradoxalement, les inhibiteurs de l’IDO inhibent eux aussi la production de cytokines M1/M2. Discussion Nous avons pu verifier que la voie des kynurenines est exprimee par les macrophages pulmonaires humains et que les kynurenines modulent leur fonction. L’effet paradoxal des inhibiteurs de l’IDO pourrait etre explique par une deviation du tryptophane au profit de la voie de la serotonine, elle-meme immunomodulatrice. Conclusion Les kynurenines (Tranilast®, 3OHAA et 3OHKYN), en agissant sur la polarisation des macrophages pulmonaires humains, participent a la regulation du systeme immunitaire pulmonaire et pourraient etre une piste de therapie dans les maladies inflammatoires chroniques comme la BPCO ou l’asthme.
