Abstract

1. Es un “retrato dinamico” del estado metabolico de los sistemas vivientes2. El metaboloma se compone de una amplia cantidad de metabolitos constituidos por pequenas moleculas y productos de los procesos celulares, que exhiben una diversidad de propiedades fisicas y quimicas y que existen dentro de un intervalo dinamico muy amplio en muestras biologicas de celulas, tejidos, organos y organismos3-5. Esta novedosa tecnica se fundamenta en el uso de la espectrometria de masas para investigar los mecanismos bioquimicos relacionados con procesos naturales en la salud y tologicas no solo se caracterizan por las identidades y concentraciones de metabolitos presentes, sino por la localizacion de estos en un tejido gracias a la cromatografia de gases o la separacion por cromatografia de liquidos de alta resolucion (HPLC) acoplados a un espectrometro de masas de velocidad y precision4. Infortunadamente, la mayoria de las plataformas de espectrometria de masas solo puede medir muestras en solucion, por lo que se requiere extraer metabolitos de tejidos homogeneizados. Sin embargo, los mas recientes desarrollos en tecnologias de espectrometria de masas-imagen permiten que ciertos metabolitos puedan reconocerse de manera espacial dentro de sus tejidos biologicos6. A traves de la caracterizacion de rutas metabolicas, la metabolomica puede ilustrar las actividades de una celula a nivel funcional. Sin embargo, el metaboloma es dificil de caracterizar y por ello la espectrometria de masas, acoplada a diferentes tecnicas de separacion, es tan util7. La proteomica consiste en el analisis profundo de la estructura y funciones del complejo de proteinas en una celula, tejido o liquido biologico en un cierto momento de la expresion de proteinas, y es una plataforma central en la nutrigenomica que describe la expresion del genoma como respuesta a la dieta. La proteomica permite entender las funciones de organelos y sus alteraciones, entre otros muchos aspectos8. En particular, la disfuncion y mutaciones del DNA explican un gran numero de enfermedades, incluido el cancer, y la proteomica es especialmente util para identicion y muerte celulares. La bondad de la proteomica y metabolomica radica en su nula invasividad (se fundamenta en el analisis de muestras biologicas facilmente obtenibles como orina, sangre y saliva) a cambio de obtener amplia informacion9.
