Abstract A technique has been developed for the in vitro demonstration of anti-nuclear antibodies (ANA) in the sera of patients with systemic lupus erythematosus (SLE) utilizing human metaphase chromosomes in situ as substrate. The selective removal of histone, acidic protein, RNA and treatment with 2% formamide improves the binding of antibodies to DNA as demonstrated by fluorescein-conjugated and horseradish peroxidase-conjugated goat anti-human IgG. This technique is not only important because of the relative ease of demonstration of ANA but may be a valuable tool in the investigation of the pathogenesis of SLE.
