RAPD markers and genetic diversity inPelteobagrus fulvidraco

Abstract

primers whose RAPD reactions are reproductive and with clean bands are selected from 422 primers by using the DNA pools of both male and female Pelteobagrus fulvidraco as templates. These 61 primers can be used as molecular marker for the species identification. We apply 10\|11 primers to proceed RAPD analysis consequently to two groups. In the first group, 95 sites (bands) are revealed, 27 of which are polymorplic, upon 28.42%; the NeiLi's genetic similarity within population is 90.67% and Shannon's information index is 0.091 8 bit. In the second group, 46 sites(bands) are revealed, 12 of which are polymorphic, upon 26.09%; the Nei Li's genetic similarity is 93.27% and Shannon's information index is 0.067 0 bit. The genetic diversity of Pelteabagrus fulvidraco is lower than that of reported Carassius auratus L. and Gobiocypris rarus and higher than Aristichthys nobilis .
