Quantitation of the effect of NH4Cl on viral penetration was done under conditions of single-cycle replication in a system using Krebs-2 ascites cells and influenza virus, which produces only unassembled viral structural protein. Penetration was measured by neutralization of adsorbed virus in the presence or absence of NH4Cl and comparison of yields of hemagglutinins after a 24 hour growth period.
