Pigeon globin mRNA isolated from reticulocyte polysomes was copied by means of RNA-directed DNA-polymerase in the presence of actinomycin d. Hydroxyapatite chromatography with and without formaldehyde and hydrolysis with S1 nuclease were used for evaluation of the secondary structure elements in cDNA synthesized by reverse transcription.
