
The BRAF/MEK inhibitors, encorafenib and binimetinib, is the FDA/EMA-approved combination used in BRAF-positive melanoma. The phase III study showed an improvement in progression-free survival (PFS) and overall survival (OS) with good tolerability. A direct comparison of monotherapy showed better results in the arm with encorafenib over vemurafenib. This review provides an overview of pharmacological properties, efficacy, and toxicity of encorafenib and binimetinib.
