All human diseases, including those of the nervous system, are the result of interactions between genetic factors and the environment. In some diseases the genetic factors are relatively minor and of little importance. Other disorders are strictly inherited conditions, and genetic factors are paramount. The major categories of genetic influence are single gene (mendelian), gross chromosomal aberration, mitochondrial, and multifactorial/polygenic. Examples of neurologic diseases representing each of these categories are illustrated in this chapter.
