Purine nucleoside phosphorylase (PNPase, purine nucleoside: orthophosphate ribosyltransferase E.C. 2.4.2.1) in mammals reversibly catalyzes the phosphorolysis of the purine ribonucleosides (deoxy)inosine and (deoxy)guanosine to (deoxy)ribose-l-phosphate and the respective base. This enzyme has been isolated and purified from various cytosolic sources (1–4). This report describes the partial characterization of a newly discovered PNPase activity isolated from the mitochondria of bovine liver.
