DNA supercoiling induced by a synthetic polypeptide

Abstract

The synthetic protein (Leu67-Lys33)100-Orn20 bound irreversibly to pBR322 circular DNA and induced supercoiling in the DNA. The no. of topoisomers reached an optimum corresponding to a wt.-wt. ratio of 1, then remained approx. const. The presence of a population of different topoisomers probably indicates nonregularity in the topol. effects caused by the nonuniformity of protein compn. and structure. A comparison between the effects on supercoiled and relaxed DNA shows that the protein binds most easily to supercoiled DNA. Two-dimensional electrophoresis in the presence of ethidium bromide indicates that the protein induces a left-handed supercoiling in DNA.
