AbstractX‐ray crystallography is one of the major tools available for protein structural analysis. This unit provides an introductory review of the principles of X‐ray crystallography that covers how the image is generated and analyzed. The second half of the unit describes strategies for producing crystals of protein, including methods for dealing with proteins that do not easily form crystals.
