THE ISOLATION AND IDENTIFICATION OF CARBOXYESTERASES OF DROSPHILA MELANOGASTER

Abstract

The isolation of the main carboxyesterases of drosophila’s imago was conducted with the help of gel filtration chromatography on sefadex G-50 and ion exchange chromatography on DEAE-sefadex. The albuminous components of initial extracts and chromatographic fractions were divided by the method of polyacrilamide gel electrophoresis. The electrophoretic fractions of drosophila’s carboxyesterases were identified on the stages of their isolation by means of carrying out the reaction of nitric combination. The acid nature of the main forms of carboxyesterases was established, their substrate specificity was revealed and the scheme of isolation and identification of these ferments was worked out.

С помощью гель-фильтрации на сефадексе G-50 и ионообменной хроматографии на ДЭАЭ-сефадексе проводили выделение основных карбоксиэстераз имаго дрозофилы. Методом электрофореза в полиакриламидном геле осуществляли разделение белковых компонентов исходных экстрактов и хроматографических фракций. Путём проведения реакции азосочетания идентифицировали электрофоретические фракции карбоксиэстераз дрозофилы на этапах их выделения. Установлена кислая природа основных форм карбоксиэстераз, показана их субстратная специфичность, разработана схема выделения и идентификации указанных ферментов.

За допомогою гель-фільтрації на сефадексі G-50 та іонообмінної хроматографії на ДЕАЕ-сефадексі провадили виділення основних карбоксиестераз імаго дрозофіли. Методом електрофорезу в поліакриламідному гелі розділяли білкові компоненти вихідних екстрактів та хроматографічних фракцій. Шляхом проведення реакції азосполучення ідентифікували електрофоретичні фракції карбоксиестераз дрозофіли на етапах їх виділення. Встановлена кисла природа основних форм карбоксиестераз, показана їх субстратна специфічність, розроблено схему виділення та ідентифікації даних ферментів.
