Genetic characterization of Rhizobia isolated from Lotus corniculatus L.

Abstract

Trinta e três isolados de Rhizobium sp obtidos a partir de nódulos de cornichão (Lotus corniculatus L.), coletados em diferentes regiões do Rio Grande do Sul, foram caracterizados pela técnica da PCR (reação em cadeia da polimerase), utilizando oligonucleotídeos específicos para a amplificação de seqüências repetitivas Box. Os isolados apresentaram perfis de amplificação distintos entre si e das estirpes de Mesorhizobium loti, SEMIA 806 e SEMIA 816, atualmente recomendadas para a produção de inoculantes para cornichão. O índice de similaridade genética determinado pelo coeficiente de Jaccard variou entre 0,5 e 0,9% indicando uma grande variabilidade entre os isolados. A análise de agrupamento dos isolados demonstrou a formação de três grupos, não tendo sido possível estabelecer relação entre o perfil de amplificação, grupos formados e origem geográfica dos isolados.

Thirty three Rhizobium sp isolates obtained from birdsfoot trefoil (Lotus corniculatus L.) collected on different regions of Rio Grande do Sul were characterized using Box-PCR. The isolates presented distinct amplification profiles among then, as well as from the isolates recommended nowadays for the production of birdsfoot trefoil inoculants, SEMIA 806 and SEMIA 816. The genetic similarity index, determined by the Jaccard coefficient, ranged from 0.5 to 0.9%, pointing out to a large genetic variability among the isolates. The cluster analysis isolates showed a formation of tree large groups and was not possible to establish a relationship among the amplification profiles, groups formed and geographic origin of the isolates.
