Изменение электрокинетических свойств эритроцитов при их консервации

Abstract

Цель исследования изучение влияния консервации эритроцитов на электрофоретическую подвижность (ЭФП) и морфологию красных клеток крови. Эритроциты выделяли из крови 30 доноров, заготовленной в консерванте ЦФДА-1. ЭФП изучали с помощью светового микроскопа в специальной камере, в которую помещали Ag/AgCl-электроды. Морфологию эритроцитов изучали с помощью светового микроскопа Primo Star («Carl Zeiss», Германия). Микровезикулы (МВ) выделяли из суспензии эритроцитов с помощью ультрацентрифугирования при 100 000 g. Установлено, что при консервации эритроцитов происходит значительное снижение их ЭФП. Одной из важных причин этого является образование МВ, связанное, в том числе, с диск-сферической трансформацией эритроцитов: развивается выраженный анизоцитоз и пойкилоцитоз. Более быстрое и значительное изменение этих процессов происходит в предварительно отмытых эритроцитах, несмотря на то, что они также хранились в консервирующем растворе. Чем сильнее и быстрее происходят морфологические изменения эритроцитов, тем более выражено снижение их ЭФП и образование МВ.

The effects of erythrocyte preservation on their electrophoretic mobility (EPM) and morphology were studied. Erythrocytes were isolated from the blood, collected in CPDA-1 preserving agent, in 30 donors. Electrophoretic mobility was studied under a microscope in a special box with Ag/AgCl electrodes. The morphology of erythrocytes was studied under a Primo Star microscope (Carl Zeiss, Germany). The microvesicles (MV) were isolated from erythrocyte suspension by ultracentrifugation at 100 000 g. The erythrocyte EPM reduced significantly after preservation. One of the causes of this reduction was MV formation, caused, among other things, by disk-spheric transformation of erythrocytes marked anisocytosis and poikilocytosis. These processes were more rapid in pre-washed erythrocytes, despite their storage in the same preserving solution. More intense and rapid morphological changes in erythrocytes were associated with greater reduction of their EPM and formation of MV.
