Abstract

Novel amperometric biosensors based on screen-printed graphite electrodes modified with multi-walled carbon nanotubes (MWCNTs), gold nanoparticles (NP Au), and the immobilized enzyme L-cysteindesulfhydrase are proposed for the determination of antidepressants (moclobemide, imipramine). The response of the proposed biosensor is based on a combination of enzymatic and electrochemical reactions and the inhibition effect of the antidepressants under study. Various methods for the synthesis of gold nanoparticles are proposed: in MWCNTs suspension and by electrochemical deposition. The biosensors can be used to control both the traces of these pharmaceuticals in biological fluids and the content of the active substance in the pharmaceutical formulations. The cysteindesulfhydrase biosensors based on modified MWCNT / NP Au electrodes allow determination of antidepressants within the linear concentration range of 1·10 –4 – 1·10 –9 mol/l. The lower limits of detection are 6·10 –10 and 9·10 –10 mol/l for moclobemide and imipramine, respectively.

Разработаны новые ингибиторные амперометрические биосенсоры на основе планарных печатных графитовых электродов, модифицированных многослойными углеродными нанотрубками (МУНТ), наночастицами золота (НЧ Au) и иммобилизованным ферментом L-цистеиндесульфгидразой для определения антидепрессантов (моклобемид, имипрамин). Предложены различные способы получения НЧ Au (в суспензии МУНТ и электрохимическое осаждение). Биосенсоры могут быть использованы для контроля как остаточных количеств антидепрессантов в биологических жидкостях, так и основного вещества в лекарственных формах в области концентраций 1·10 –4 ÷ 1·10 –9 моль/л. Нижняя граница определяемых содержаний на уровне 6·10 –10 и 9·10 –10 моль/л для моклобемида и имипрамина соответственно.
