Abstract

The aim of the research. To study the regulatory role of connexin 43 (Cx43) in intercellular interactions in the neurovascular unit (NVU) in vitro. Materials and methods. The research was conducted on a model NVU in vitro with the evaluation of Cx43 and CD38 expression in physiological conditions and in the suppression of Cx43 expression. Results. NVU cells-components in vitro express Cx43 and CD38. Application of siRNA reduces the expression of Cx43 and CD38 on neurons and endotheliocytes. There is a direct correlation of medium strength between expression of Cx43 and CD38. Conclusion. During the work it was established that there is functional conjugation between the molecule CD38 and Cx43, distinguished by their characteristics in NVU cells and indicated of their different sensitivity to the action of factors that violate the metabolism of NAD +.

Цель исследования. Изучение регулирующей роли коннексина 43 (Сх43) в межклеточных взаимодействиях в нейроваскулярной единице (НВЕ) in vitro. Матери алы и методы. Исследование проводилось на модели НВЕ in vitro с оценкой экспрессии Сх43 и CD38 в физиологических условиях и при подавлении экспрессии Сх43. Результаты. Клетки-компоненты НВЕ in vitro экспрессируют Cx43 и CD38. Аппликация siRNA приводит к снижению экспрессии Сх43 и CD38 на нейронах и эндотелиоцитах. Существует прямая корреляционная связь средней силы между экспрессией Сх43 и CD38. Заключение. В ходе работы установлено, что существует функциональное сопряжение между молекулой CD38 и Сх43, различающееся по своим характеристикам в клетках НВЕ и свидетельствующее об их разной чувствительности к действию факторов, нарушающих метаболизм НАД+.
