«Постгеномный взгляд» на проблемы онкогенеза

Abstract

Processes of malignant transformation of cells and the subsequent tumoral progression, possibly, are a consequence of common reason the genetic instability arising at damaging one of the genes of the system of genomic self-checking («stability genes»). Mutations in these genes provide considerable decrease in efficiency of revealing and a reparation of the subsequent genetic damages and errors of DNA replication. Descendants of such mutant cell reproduce a tolerance phenotype to new genetic damages owing to what rate of accumulation of the mutations changing a phenotype and breaking gene programs sharply increases in them. Against mass accumulation of every cell of a casual set of mutations malignant transformation one or several of them is possible. As a result of the same process of the further accumulation of mutations initially monoclonal tumour is subject to clonal splitting and a progression.

прогрессии, вероятно, являются следствием общей причины генетической нестабильности, возникающей при повреждении одного из генов системы самоконтроля генома («генов стабильности»). Инактивирующие мутации в этих генах ведут к значительному снижению эффективности выявления и репарации последующих генетических повреждений и ошибок репликации ДНК. Потомки такой мутантной клетки воспроизводят фенотип толерантности к новым генетическим повреждениям (мутаторный фенотип), вследствие чего в них резко возрастает скорость накопления мутаций, изменяющих фенотип и дискоординирующих генные программы. На фоне массового накопления каждой гипермутабельной клеткой случайного набора мутаций возможна опухолевая трансформация одной или нескольких из них. В результате того же процесса дальнейшего накопления мутаций первично возникшая моноклональная опухоль подвержена клональному расщеплению и прогрессии.
