Биохимические механизмы формирования тромбофилии, индуцированной хронической нагрузкой тиолактоном гомоцистеина и его комбинацией с L-NAME. Коррекция витаминно-микроэлементным комплексом

Abstract

Исследовано вплияние хронической нагрузки тиолактоном гомоцистеина (100 мг/ кг) и его комбинации с L-NAME (30 мг/кг) на систему гемостаза крыс. Нагрузка тиолактоном гомоцистеина вызывает комплексные нарушения в системе гемостаза: гиперреактивность тромбоцитов, тромбинемию, гиперфибриногенемию, снижение антикоагулянтной и фибри-нолитической активности. Установлено, что новым паттерном формирования тромбофилии, индуцированной гипергомоцистеинемией, является дефицит вазодилятатора, антиагреганта и антиоксиданта гидроген сульфида. Введение L-NAME усиливает дефицит гидроген сульфида и потенциирует негативное влияние тиолактона гомоцистеина на систему гемостаза крыс. Обогащение рациона крыс витаминно-микроэлементным комплексом уменьшает дефицит гидроген сульфида и препятствует развитию тромбофилии.

It was investigated influence of homocysteine thiolactone loading (100 mg/kg) and its combination with L-NAME (30 mg/kg) within 4 weeks on rat hemostasis system. Homocysteine thiolactone loading caused complex disturbance in hemostasis system such as platelet hyperreactivity, thrombinemia, hyperfibrinogenemia, decreasing of anticoagulant and fibrino-lytic activity. It was established that deficit of vasodilator, antiaggregant and antioxidant hydrogen sulfide became the new pattern of thrombophilia formation induced by hyperhomocys-teinemia. L-NAME administration increased hydrogen sulfide deficit and potentiated negative influence on rat hemostasis system. Vitamins-microelement enrichment of rat diet decreased hydrogen sulfide deficit and prevented from thrombophilia formation.
