Экспрессия генов раково-тестикулярных антигенов в первичной меланоме кожи человека

Abstract

Методом полимеразной цепной реакции, совмещенной с обратной транскрипцией, выявляли корреляцию активности 13 генов раково-тестикулярных антигенов, 4 дифференцировочных антигенов меланоцитов и 14 генов маркеров меланомы в 26 образцах первичной меланомы человека с митотическим индексом клеток первичной меланомы и прогнозом заболевания. Показано, что анализ экспрессии 32 генов позволяет разделить всех пациентов на 3 группы: с низкой и высокой 5-летней выживаемостью при высокой доле пациентов с метастазами и пациентов без признаков заболевания. Генная экспрессия выбранных для исследования генов может использоваться как независимый прогностический фактор продолжительности жизни пациента после хирургической операции и склонности к метастазированию с высокой специфичностью и чувствительностью.

Gene expression of 13 cancer/testis antigens, 4 melanocytic melanoma differentiation antigens and 14 melanoma markers was analyzed with respect to to mitotic index of primary melanoma cells by RT-PCR in 26 human primary melanoma specimens. Basing of expression of 32 genes all patients were divided into 3 groups of low 5-year survival, high 5-year survival with a large fraction of patients having metastatic disease, and apparently disease-free patients. Expression of the selected genes can be used as independent, highly specific and sensitive test for prognosis of survival and disease metastatic potential in patients undergoing surgery.
