Abstract

Co-cultivation of human thymocytes and thymotropic Acanthamoeba cells (the CDHT strain) for 24 hours resulted in a significant change of the relative number of thymocytes representing different subpopulations. The most pronounced was the change in the proportion of immature cortical thymocytes with the CD4 + CD8 + membrane phenotype. Simultaneously, the percentage of the least and most mature cells (CD4 + CD8 and CD4 CD8 respectively) increased. The increase in the relative number of mature CD4 -CD8 + cells was much less apparent. The proportion of thymocytes with lowand high-intensity expression of CD3 (mature and immature ones respectively) tended to change in a similar manner, that is the number of CD3 lo decreased while that of CD3 hi increased. The percentage of CD4 + CD25 hi thymocytes (i.e. natural regulatory T-cells) increased under the influence of co-cultivation with the Acanthamoeba cells. At the same time, the relative number of CD4 + CD25 lo cells (i.e. activated CD4 + thymocytes) remained unaltered. In the case of separation of the co-cultured thymocytes and CDNT cells by a semipermeable membrane or after the addition of humoral products of the Acanthamoeba cells into the culture of thymocytes, the changes in the subpolulation composition were on the whole similar to those described above but much less pronounced (the difference insignificant). Effects of co-cultivation of thymocytes with thymotropic Acanthamoeba cells were analogous to the effects of any cytotoxic agents including glucocorticoids and ionizing radiation, It is hypothesized that all the observed changes are associated with the induction of apoptosis to which CD4 + CD8 + thymocytes show the maximum sensitivity.

При совместном культивировании тимоцитов человека с клетками акантамебы, тропной к тимусу (штамм CDHT), в течение 1 сут существенно изменяется численное соотношение тимоцитов, относящихся к различным субпопуляциям. Основной сдвиг состоит в снижении относительного содержания незрелых кортикальных тимоцитов с мембранным фенотипом CD4 +CD8 +. Одновременно происходит увеличение процента наименее зрелых (CD4 -CD8 -) и наиболее зрелых (CD4+CD8 -, в значительно меньшей степени CD4 -CD8 +) клеток. Соотношение тимоцитов, экспрессирующих слабо и сильно CD3 соответственно незрелых и зрелых тимоцитов изменяется аналогичным образом: снижается содержание CD3 lo-клеток, и возрастает уровень CD3 hi-клеток. Процентное содержание CD4 +CD25 hi-тимоцитов, т. е. естественных регуляторных Т-клеток, повышается под влиянием сокультивирования с клетками амебы, тогда как процент CD4 +CD25 lo-тимоцитов, т. е. активированных CD4 +-тимоцитов, при этом не изменяется. При разделении сокультивируемых тимоцитов и клеток CDHT, разделенных полупроницаемой мембраной, а также введении в культуру тимоцитов гуморальных продуктов амебы наблюдаются субпопуляционные сдвиги той же направленности, но выраженные значительно слабее (незначимо). Эффекты сокультивирования с амебоцитами аналогичны действию любых цитотоксических агентов, включая глюкокортикоиды и ионизирующую радиацию. Постулируется, что во всех этих случаях основой наблюдаемых изменений является индукция апоптоза, к которому наиболее чувствительны СD4 +СD8 +-тимоциты.
