PML-зависимые механизмы репарации повреждений дНк перспективы создания нового метода лабораторной диагностики для оценки химиои радиочувствительности злокачественных новообразований

Abstract

Было выявлено, что в условиях нокдауна белка PML на фоне повреждений ДНК, вызываемых химиопрепаратами (доксорубицин, этопозид и др.) и ионизирующим излучением, происходит нарушение фокального распределения белков, участвующих в репарации повреждений ДНК по механизму гомологичной рекомбинации. Данный факт позволяет рассматривать злокачественные новообразования со сниженной экспрессией белка PML как потенциально восприимчивые к проведению химиои радиотерапии. Разработка метода лабораторной диагностики химиои радиочувствительности злокачественных новообразований, основанная на оценке способности опухолевых клеток репарировать повреждения ДНК, вызываемых химиопрепаратами и ионизирующим излучением, представляется перспективной и позволит обеспечить правильный выбор тактики при проведении персонифицированной терапии онкологических больных, а также проведения последующего мониторинга ее эффективности.

It was found that in terms of PML protein knockdown against the background of DNA damage, caused by chemotherapeutic agents (doxorubicin, etoposide and etc.) and radiation, happens abnormality of focal distribution of proteins which participate in DNA damages reparation by a mechanism of homologous recombination. This allows us to consider malignant neoplasms with reduced PML protein expression as potentially susceptible to chemotherapy and radiotherapy. Development of laboratory diagnostic method of chemo-and radiosensitivity of malignant tumors, based on an assessment of the ability of tumor cells to repair DNA damage, caused by chemotherapy and ionizing radiation, seems promising and will provide the right choice of tactics in the process of the patient-specific therapy of oncologic patients, as well as follow-up monitoring of its effectiveness.
