Abstract

Разработан лабораторный метод выделения концентрата фактора IX (ФIХ) высокой степени очистки, включающий последовательные ступени ионообменной, аффинной и металл-хелатной хроматографии. Оптимизированные условия проведения стадий аффинной и металл-хелатной хроматографии позволяют обеспечить выход фактора свертывания крови IX (ФIХ) не менее 83 ± 10 и 82 ± 7% соответственно. Получаемый концентрат с удельной активностью ФIХ 19,6 ± 5,5 МЕ/мг содержит не более 0,0001; 0 и 0,0004 МЕ ФII, ФVII и ФХ на 1 МЕ ФIХ. Полученные результаты могут быть положены в основу технологии получения антигемофильного препарата ФIХ высокой степени очистки из плазмы донорской крови.

The new laboratory method for isolation of highly purified concentrated factor IX (FIX) includes a series of stages ion exchange, affinity, and immobilized metal ion affinity chromatographies. Optimal conditions for affinity and immobilized metal ion affinity chromatography have resulted in FIX output of 83±10 and 82±7%, respectively. The resultant concentrate with FIX specific activity of 19.6±5.5 U/ml contains no more than 0.0001, 0, and 0.0004 U FII, FVII, and FX per unit of FIX. The results can be used for developing a technology for obtaining highly purified concentrated FIX from human plasma.
