Abstract

Причиной развития ОПЛ является появление химерного онкогена PML/RARα, экспрессия которого наблюдается при данном заболевании во всех опухолевых клетках и используется в качестве молекулярного маркера для установления диагноза и определении минимальной остаточной болезни. Кроме того, при ОПЛ, так же, как и при многих других злокачественных новообразованиях, экспрессируется неспецифический онкомаркер PRAME, относящийся к группе канцер-тестис антигенов. В настоящей работе экспрессия гена PRAME нами исследовалась у больных ОПЛ в сопоставлении с уровнем экспрессии гена PML/RARα, а также с данными о клиническом течении этого заболевания. Выяснилось, что в дебюте ОПЛ обязательно наблюдается коэкспрессия генов PRAME и PML/RARα. По отношению уровня экспрессии гена PRAME к PML/RARα в дебюте заболевания пациентов с ОПЛ можно разделить на две группы - с высоким и низким отношением PRAME к PML/RARα. Больные ОПЛ, имевшие в дебюте низкое соотношение PRAME к PML/RARα, были склонны к более раннему развитию рецидива, чем лица с высоким соотношением уровней экспрессии данных генов.

An appearance of a chimeric oncogene PML/RARα is a reason of an APL. This molecular marker is used for diagnosis and minimal residual disease monitoring in patients with APL. PRAME gene related to a group of cancer-testis genes is expressed in APL and a lot of other malignancies. We studied the ratio between expression levels of genes PRAME and PML/RARα and compared it with a clinical data of patients with APL. According to this dividing there were two groups of APL patients: with a high and low ratio respectively. Furthermore patients with a low ratio had had a leukemia relapse earlier than patients with the high ratio values. We suggest that the ratio between their expression levels accurately determines a risk of relapse.
