Abstract

The aim of the investigation is to give comparative assessment of melatonin (Melaxen) and the derivative of 3-hydroxypyridine (Mexidol) effect on hematotoxicity and antitumour efficacy of alkylating cytostatic Cyclophosphan in mice with Lewis lung carcinoma. Materials and Methods. The experiments were carried out on 80 female mice of C57Bl/6 line, weighing 20-22 g. Cyclophosphan was administered twice at the dosage of 100 mg/kg at 96h intervals intraperitoneally, and Melaxen and Mexidol intramuscularly at the dosages of 50 and 45 mg/kg respectively, within 14 days. In each group of animals there was assessed hematotoxicity, treatment efficacy by the index of tumour growth inhibition, and antimetastasis effect by an average number of superficial lung metastases per an animal. Conclusion. Melaxen and Mexidol decrease Cyclophosphan hematotoxicity without reducing its antitumour efficacy in regard to primary tumour node. Cyclophosphan and Melaxen used in combination do not change antimetastasis effect, while the latter is increasing when cytostatic agent is used with Mexidol compared to the administration of Cyclophosphan only.

Цель исследования сравнительная оценка влияния мелатонина (Мелаксена) и производного 3-гидроксипиридина (Мексидола) на гематотоксичность и противоопухолевую эффективность алкилирующего цитостатика Циклофосфана у мышей с карциномой легкого Льюис. Материалы и методы. Эксперименты выполнены на 80 мышах-самках линии C57Bl/6 массой 20-22 г. Циклофосфан вводили в дозе 100 мг/кг двукратно с интервалом 96 ч внутрибрюшинно, Мексидол и Мелаксен внутримышечно в дозах 50 и 45 мг/кг соответственно, в течение 14 дней. Оценивали гематотоксичность у животных каждой группы, эффективность лечения по индексу торможения роста опухоли, антиметастатический эффект по среднему числу поверхностных легочных метастазов на одно животное. Заключение. Мелаксен и Мексидол снижают гематотоксичность Циклофосфана, не уменьшая его противоопухолевой эффективности в отношении первичного опухолевого узла. Антиметастатический эффект при сочетанном применении Циклофосфана и Мелаксена не изменяется, а при использовании цитостатика с Мексидолом увеличивается по сравнению с введением одного Циклофосфана.
