Abstract

In this work we described organization of homologous chromosomes in interphase nuclei of nerve cells of the human brain. The cortex and cerebellum of postmortem brain in normal and ataxia-teleangiektasia (AT) a genetic disorder, characterized by chromosomal instability and progressive cerebellar degeneration, were analyzed by FISH. It was shown that the normal and pathological brain cells characterized by the phenomenon of somatic pairing (or association) of chromosomal loci that affect the heterochromatic regions of different chromosomes. This finding indicates for the violation of the rules of «chromosome territories» in the brain cells describing the specific positioning of homologous chromosomes in the nucleus. We have determined the frequency of somatic pairing of heterochromatic regions of different human chromosomes in the cells of the cortex and cerebellum in the normal and AT brain. We have detected the reduced rate of chromosome 7 pairing the in the AT cerebellum compared with the control, which may be associated with cerebellar neuronal loss and instability of certain chromosomal loci in the cells of the degenerating cerebellum in AT.

В работе охарактеризованы особенности организации гомологичных хромосом в интерфазных ядрах нервных клеток коры и мозжечка головного мозга человека в норме и при атаксии телеангиэктазии (АТ) – генетическом заболевании, для которого характерна хромосомная нестабильность и прогрессирующая мозжечковая дегенерация. Показано, что в клетках мозга в норме и при патологии наблюдается феномен соматического спаривания (или ассоциации) хромосомных локусов, затрагивающих гетерохроматиновые участки разных хромосом, что свидетельствует о нарушении правила «хромосомных территорий» для гомологичных хромосом в клетках мозга. Впервые определены частоты соматического спаривания гетерохроматиновых районов разных хромосом человека в клетках коры и мозжечка головного мозга в норме и при АТ. Выявлены различия в частотах ассоциаций хромосомы 7 в мозжечке при АТ по сравнению с контролем, которые могут быть ассоциированы с нейрональной гибелью и нестабильностью определенных хромосомных локусов в клетках мозжечка при АТ.
